Reading the bitterness
Cocoa powder consists of defatted and ground roasted cocoa solids and is distributed as both natural product and product subjected to alkalization (the Dutch process). A study comparing before and after microwave-assisted alkalization chemically analyzed combinations of constituents responsible for bitterness and astringency.
In the same study’s Dose-Over-Threshold analysis, epicatechin, (-)-epigallocatechin gallate, luteolin, protocatechuic acid, theobromine, and caffeine were judged to contribute significantly to cocoa-powder bitterness (DOT>1). The constituents responsible for astringency partly differed, with syringic acid, p-coumaric acid, and 5-hydroxytryptamine among those playing important roles. Methylxanthines such as theobromine and caffeine appeared in this set alongside phenolic compounds, so they are not the sole bitterness-driving factors in this material.
Alkalization is positioned as an important cocoa-processing step for reducing bitterness, increasing solubility, and developing color. In a test applying different alkalization conditions for commercial cocoa powder to sponge cake, differences in treatment conditions affected batter properties and the cake’s color and texture, but the consumers’ overall evaluation itself was reported not to differ significantly by treatment condition.
These findings remain confirmed only at the level of abstracts or secondary summaries. Limitations include the source of the DOT-analysis thresholds and concentrations, the fact that receptor binding modes were based on molecular docking and simulation rather than human-receptor assays, and the fact that the alkalization study did not reach numerical sensory scores for bitterness itself.

Checked for this overview: Confirmed that it was analyzed for bitterness and astringency constituents before and after alkalization. / Confirmed the difference between the 6 bitterness constituents with DOT>1 and the astringency constituent set. / Confirmed the limitation that receptor analysis is limited to docking and MD.
Checked for this overview: Confirmed the role of alkalization and the results of the cake-application test.

Scope: Cocoa powder before and after microwave-assisted alkalization
Note: Methylxanthines appear in the same DOT set alongside 4 kinds of phenolics, and are not the sole bitterness driver in this material.

In Experiment 1, cocoa flavoring and vanilla flavoring were added to an instant cocoa beverage; in Experiment 2, vanilla flavoring was added to a milk beverage made bitter with caffeine. Sensory profiling was compared with and without a nose clip.
Note: The same source contains both an example that moved as expected from aroma-taste congruence (cocoa aroma toward bitterness, vanilla aroma toward sweetness) and an example that moved against that congruence (vanilla aroma strengthening the bitterness of a milk beverage). Within the scope of the abstract.

Note: External audit N2-2 (2026-10-20): re-read in the full text from the institutional repository (RiuNet, hdl 10251/189867). The statement that alkalization reduces bitterness is an introductory sentence citing a review (Valverde-Garcia et al. 2020); this study itself did not compare bitterness with non-alkalized cocoa. In its sensory evaluation (9 trained panelists, 0 to 5 scale), the more bitter samples were CM_KOH and NM_K2CO3 under mild conditions and CS_K2CO3 and CS_KOH under strong conditions, so bitterness did not decrease as the treatment got stronger. The status is preliminary until the primary basis of the review is checked.
